Single molecule manipulation make possible to disrupt molecular bonds that hold native structures in nucleic acids and proteins. In this talk I will present recent results on irreversibility and dissipation in nucleic acid hairpins that are mechanically unzipped using laser tweezers. I will focus my attention on the relation between dissipation and molecular fragility in specifically designed DNA molecules and discuss connections between energy dissipation and molecular misfolding.
